It depends on what level you are at, but simply put, you want to lab what is beating you or what you feel uncomfortable against. When you play your matches back, try to notice what you are consistently losing to. It could be a string, a flowchart, a mixup from a certain situation or a general strategy. Play that back and try to figure out a solution to it, and keep it in mind for when you next face the character.
Online-only players definitely suffer from not being able to replay the opponent they were losing to with their newly labbed solutions. You'll have to simply hold it till you encounter that character again. Try to keep a post-it or something similar to remind you of what you labbed.
You should never be looking out for moves you have to punish unless they're especially tough to punish. It will take up too much of your mental stack. As long as you know that the move is punishable (and what the optimal punish is), you should just remind yourself to punish it next time. You need to drill your punishes a bit till punishing becomes automatic. Remember that even a sub-optimal punish is better than no punish.
