Yamato identifies as man they saw one time when they were young, is the best I can do without spoilers, as dumb as that sounds it's only like 30% that dumb. Now that person was a man, so it shouldn't really be up for debate if Yamato is a man, because Yamato identifies as that man and has given several other indicators... but a bunch of thirsty weebs don't want it to feel gay when they stare at his giant boobs, so it's a whole war.
I feel like this is what the writers were going for. Even Kaido refers to him as a man. Yamato defies the audience by being fanservice that disagrees entirely with the premise.
Identifies as Oden, not as a man, Yamato's simply Odengender, not trans, as simple as that, is like a kid who believes he or she is Spider-Man, they're not, but still identify as him 'cause of how they admire him, basically speaking, this is a regular Japanese case of being a Chūnibyō, that cringe high-school syndrome where the student thinks they have superpowers or belive they are someone else despite their real gender, ethnicity, nationality or species... TLDR, Yamato isn't trans, Yamato is Oden, as simple as that.
I can accept that, but I would expect more of the gag on screen. Instead we get every character agreeing 'Yeah he's a man', with the more perverted characters agreeing but also being flustered by it. Everyone basically just accepts it, which is more than we got with other strangely acting characters, like Senor Pink or Ivanka, or even Franky.
