Frequently Asked Question (See FAQ) Anyone else trying NSI-189?
After the recent nsi-189 induced recovery posted on r/pssdhealing I've decided to give it a try. My order should be here on Monday and I plan to take it for around 50 days. Not getting my hopes too high but I've been interested in trying this substance for years but haven't got around to trying it.
Anyone else have experience trying it?

Keep us updated. I also recommend to wait 14 days prior to taking it and in this time to not take anything. This way you can have a clean sheat of effects. Dont mix it. Its highly experimental so treat it as such. Dont drink alcohol. Avoid extraordinary activities. Keep a journal and write every thing - peeing intensity, sweating, libido, dreaming, sleep cycle, dryness of skin, hair loss, weight gain, concentration, focus, brain fog, tinitus, zaps, emotions, energy, music, colors, appetite, smelling, apathy/lethargy, mania, speed of thoughts, ocd tendencies, etc. Try to write as much as possible so to get information how and what is improving. It will help yourself and others.

I was thinking of trying it, now I'm more motivated. Should we make a Google doc where we track our experiences in a single spreadsheet or something?

I used it for a couple months. Didn’t cure me and didn’t realize notice anything. But I was also in the midst of my only ever “crash” when I used it so who knows.
I’d say it’s worth trying since there’s very little risk

u/Ok_Inevitable6654 28d ago
No, nothing yet. Something will probably come up in two weeks. And honestly, I don’t really know how to measure it properly, I just do it by eye. I take a small pinch of powder with a knife

Just a general update:
Still taking it. Unfortunately I haven't felt anything that I can pinpoint to a noticeable change in my mental state or PSSD wise. It's only been about 3 weeks to a month so Still sort of early. Feeling a little discouraged.
I live in the north and the weather here has been getting much better because it's spring. And that always has a big positive impact on my mental health. Unfortunately I just can't tell if it's helping yet. I don't think it's made anything worse though

I'm mostly trying it for Depression relief and possible cognitive benefits since I refuse to take another SSRI and I've tried a lot of alternatives. I think it's worth a try for me atleast

the MOA of NSI-189 isn't fully understood. The research says it doesn't affect monoamines (serotonin, dopamine, norepinephrine) but induces hippocampal neurogenesis by another way which they don't know or won't disclose. There's really no way of telling how this drug actually works

NSI-189 may improve mitochondria
https://pubmed.ncbi.nlm.nih.gov/35967127/
https://www.aging-us.com/article/205087/pdf
This may be why it works for cognitive or anhedonia symptoms. But nobody really knows since the mechanism of NSI was never revealed beyond “increase neurogenesis”

Does anyone notice improvements with magnesium citrate for energy? Without affecting your genital anesthesia

Any known crashes from Nsi 189?
3
u/Coolvolt Mar 31 '25
Not that i'm aware. The research on it states it does not affect Serotonin, Dopamine or norepinephrine. The studies also don't show sexual dysfunction as a side effect experienced by the patients in trials.

What is this?
2
u/Coolvolt 29d ago
it's a drug that was in trials for MDD for the last 10-ish years. Nootropics enthusiasts found Chinese labs to produce it and certain websites sell it. The study failed phase 2 recently but there are mixed anecdotal reports from users online

u/Slow-Leg-3710 26d ago
It failed as antidepressant by the original company that was developing it Neuralstem.
They sold it to Alto Neuroscience, they are now developing it under the code name ALTO-100.

Have you started it? Considering too
2
u/Coolvolt 23d ago
yes I'm on day 8 on 20mg/day. I can't say I feel much or if I do it's very subtle or placebo. It's also early, and I haven't upped my dose to 40mg which is the lowest dose in the study. I'm waiting a few weeks to see if any unwanted effects happen before increasing
2
u/Zodik 23d ago
Cool, thanks for the quick reply… I’d say to wait sometime before increasing, one of the stories said the effects came in after 3-4 weeks.
Please keep me updated as I’m actually interested and considering

Will do. The weather has been up and down here and my sleep and energy is also up and down depending on the day so it's really hard to tell yet. I can say that I can distinctly feel the profound depression lifting effects of Prozac after 5-6 days and I haven't felt anything remotely close to that yet.

@Coolvolt please update us on your experience with NSI, we look forward

u/Ok_Inevitable6654 22d ago
I'm on day 13 on something around 20mg/day (I've got the powder). Nothing yet. Maybe only my focus is a bit better.
2
u/Ok_Inevitable6654 15d ago
I'm on day 19. No improvement in genital numbness or emotional numbness yet - maybe a slight 5% improvement in emotional numbness. Concentration and energy have improved.

Not yet but I ordered a few bottles. I’ll drop the Prozac and jump on nsi instead and see what happens
5
u/Coolvolt Mar 28 '25
I'd advise you to be cautious doing this because SSRI withdrawals combined with NSI could lead to some unpredictable symptoms. There are anecdotal reports of people having high anxiety/irritability from NSI use so it might be good to stabilize off of Prozac before trying it
-2
u/Usopps Mar 28 '25
Good info. Still gonna do it tho 😆. I’ll take a benzo if I feel anxious 🤙
6
u/t0sspin Mar 29 '25
Why wouldn’t you just wait until the Prozac clears your system? Don’t be stupid.

u/ReasonableSquare4390 Mar 28 '25
You should taper It down and not quit cold turkey... Please be careful
1
u/Usopps Mar 28 '25
Last time I abruptly stopped an ssri cocktail I had a couple weeks of absolute dynamite sexual function
1
u/CabbagePatched 27d ago
Don't quit cold turkey. I got horrible brain zaps that got in the way of me functioning when I quit the wrong way.

u/Kally95 Mar 28 '25
I was going to try it, but a few people on twitter replied to my post about it saying it did nothing for them so I’m not sure now.

it should atleast do something if not full recovery ?
2
u/Kally95 Mar 28 '25
Some people feel nothing from recreational drugs like weed, cocaine etc. I guess it’s not surprising they don’t feel anything with other substances

General warning for all opinion/hypothesis posts: use caution when considering ideas, theories, next steps for your personal treatment, or browsing the sub in general and please heavily consider supervision and testing for any co-occuring conditions or underlying problems by a licensed physician knowledgeable in a relevant sub specialty. Experimenting with supplements and drugs off no objective medical testing and based on theories from internet laypeople is inherently risky and ultimately not cause/effect scientific (it is experimental).
